“Interesting viewpoint:
Why did IL-6 inhibition fail in cardiovascular disease?
ZEUS provides a sobering translational lesson:
- 6,376 patients with ASCVD, CKD and inflammation
- Ziltivekimab vs placebo
- Strong inflammatory biomarker suppression
- MACE: HR 0.99 (95% CI 0.88 – 1.11)
- Signal for more serious infections
Takeaway: Target engagement is not clinical validation.
CRP reduction does not necessarily translate into plaque modification or fewer cardiovascular events.
Perhaps the bigger lesson is for genetics-guided drug development: causal biology is not equal to guaranteed therapeutic efficacy.
