Bassem M. Mohammed: How Factor VIII Interacts To Procoagulant Membranes To Support Activation Of Blood Clotting

Bassem M. Mohammed, Assistant Professor at Saint Louis University School of Medicine, shared a post on LinkedIn about a recent article he authored, published in Journal of Thrombosis and Haemostasis, adding:

Exciting News from the Lab!

I am thrilled to share that our latest manuscript, ‘Structural determination of lipid-bound Factor VIII,’ is now in press in the Journal of Thrombosis and Haemostasis (JTH)!!!

Visualizing exactly how a coagulation factor that requires shallow hydrophobic insertion and tangential polar contacts interacts with procoagulant membranes has been a persistent biophysical hurdle.

In this work, we provide a rapid and reproducible approach to overcome this constraint using readily available reagents while defining the exact mechanism of FVIII docking on lipids and shedding light on the assembly of the intrinsic tenase complex.

Using this pipeline, we resolved these membrane-bound structures at a near-atomic resolution on lipid nanodiscs and liposomes.

Grateful for the invaluable lipid work insights with Wade Zeno, and the critical MD simulation guidance with Lamees Hegazi which was instrumental in helping us interpret previous MD simulation data in the context of our new cryo-EM findings!”
