Jesmond Dalli: How Erythroblasts Shape Neutrophil Development and Function
Jesmond Dalli, Professor in Molecular Pharmacology, Sir Henry Dale Fellow, Director QMUL Lipid Mediator Unit at Queen Mary University of London, shared a post on LinkedIn about a recent article he and his colleagues co-authored, published in Blood, adding:
“How are neutrophils programmed before they ever reach the circulation?
We are pleased to share our new paper in Blood, ‘Erythroblast-derived lipid mediators program neutrophil development and function.’
Neutrophils are shaped not only by signals encountered in peripheral tissues, but also during their development in the bone marrow. In this study, we identify erythroblasts as an important local source of specialised pro-resolving mediators (SPMs), generated through Alox15-dependent pathways.
Our findings show that erythroblast-derived SPMs maintain the integrity of erythroblastic islands, support balanced neutrophil maturation and restrain excessive neutrophil activation.
Loss of this pathway led to neutropenia, altered neutrophil function, impaired bacterial clearance and more severe experimental colitis.
Reconstitution with RvD5n-3 DPA; restored some of these defects.
Together, these findings reveal an erythroblast–macrophage–neutrophil axis through which resolution biology contributes to haematopoietic homeostasis.
We are very grateful to our collaborators and co-authors for their outstanding contributions and the funders Barts Charity for their support.
We are also honoured that Blood selected an image from the study for the issue cover.
My sincere thanks to Oliver Soehnlein and Vanessa Suerdieck for their insightful accompanying commentary, which places these findings in the broader context of bone-marrow regulation of neutrophil production.”
