Mehran Ghasemzadeh: Microparticle-Associated Mitochondrial DNA in Stored Platelet Products
Mehran Ghasemzadeh, Professor at High Institute for Research and Education in Transfusion Medicine, shared on LinkedIn about a recent article he and his colleagues co-authored, published in Experimental and Molecular Pathology, adding:
”Platelet-derived mtDNA in stored platelet products: a potential inflammatory signal or sequestered cargo within a carrier with pleiotropic biological effects?
I am pleased to share our recent publication in ‘Experimental and Molecular Pathology’, made possible by the collaborative efforts of our dedicated research team. I am particularly pleased to acknowledge my talented Master’s student, Zeinab Nazari Robati, for her first valuable contribution to scientific research through this collaborative work.
Our paper:
Partitioning of platelet-derived mitochondrial DNA between microparticle (MP)-associated and MP/platelet-depleted fractions during apheresis platelet storage: Predominance of a conserved MP-associated fraction.
Key finding: During apheresis platelet storage, mtDNA levels in the MP-enriched fraction were approximately 13-fold higher than those detected in the corresponding MP/platelet-depleted supernatant.
Why does this matter?
Unlike exposed mtDNA, which can activate innate immunity as a damage-associated molecular pattern (DAMP), MP-associated mtDNA may be less accessible to extracellular degradation and immune recognition. Moreover, platelet MPs carry diverse bioactive cargoes, potentially mediating pleiotropic effects beyond mtDNA-driven inflammation.
Our findings highlight the importance of distinguishing extracellular mtDNA fractions when interpreting platelet-storage lesions and their potential biological consequences. The functional significance of these distinct fractions warrants further investigation.
My sincere appreciation to all co-authors for their contributions to this collaborative work.”
