Axel Diederichsen: Can Vitamin K2 and Vitamin D3 Slow the Progression of Severe Coronary Artery Calcification?

Axel Diederichsen, Professor at OUH Odense Universitets Hospital, shared a post on LinkedIn:

”Can vitamin K2 and vitamin D3 slow the progression of severe coronary artery calcification?

Today at ESC Congress, we present the results of the randomized, double-blind DANCODE trial.

398 participants with severe coronary artery calcification (CAC of 400 or greater) were randomized to vitamin K2 (720 μg/day) plus vitamin D3 (25 μg/day) or placebo for 24 months.

CAC increased by 196 AU with vitamin K2 plus D3 versus 248 AU with placebo — a between-group difference of −52 AU (95 percent CI, −79 to −24), corresponding to approximately 21 percent less CAC progression.

The treatment effect was consistent across sex and baseline CAC subgroups.

Importantly, in the CCTA substudy, the lower progression of calcified plaque was not accompanied by an increase in noncalcified plaque volume.

These findings suggest that targeting vitamin K–dependent calcification pathways may slow progression of severe coronary calcification. Whether this translates into fewer cardiovascular events remains to be determined.
