Ralf Ludwig, Director of the Lübeck Institute of Experimental Dermatology at the University of Lübeck, shared a post on LinkedIn:
”How do you test a human-targeted therapy on a human immune system — before the first patient?
In this study we used humanized immune system (HIS) mice: animals reconstituted with human blood cells, carrying human leukocytes and human Fcγ receptors.
We induced an EBA-like skin disease and treated with the BTK inhibitor ibrutinib — and disease was significantly reduced, driven by human cells.
The model itself was established by Anja Lux and colleagues within CRC 1526, and described here: ‘A human immune system mouse model for preclinical evaluation of therapies in pemphigoid disease‘(Voss et al.)
It’s the bridge most autoimmune research still lacks: a place to ask whether a therapy actually touches human immunity in vivo — not just mouse.
We used it to test the impact of kinase inhibitors on pemphigoid diseases.”
