“Platelet Estimation Pitfalls: Small Details, Significant Consequences
The Barbara Brown method is useful for platelet estimation, but several factors can compromise accuracy:
- Platelet clumping and satellitism.
- Giant or small platelets.
- Uneven platelet distribution.
- Poor smear quality or staining.
- An unvalidated multiplication factor.
How can we improve accuracy?
- Examine the entire smear and select a representative monolayer.
- Standardize field selection and counting procedures.
- Verify the multiplication factor locally.
- Correlate smear findings with analyzer results and clinical information.
Remember: Microscopic platelet estimation requires more than counting. Morphology, methodology, and clinical correlation matter.
