Alan Nurden: Comparing Antiplatelet and Antithrombin Therapies in Coronary Microcirculation
Alan Nurden, Emeritus Research Director at CNRS, Co-Founder of the French National Reference Centre for Inherited Platelet Disorders (CRPP), shared a post on LinkedIn about a recent article by Joanne L. Mitchell et al., published in Basic Research in Cardiology, adding:
“This provocative manuscript looks in depth at the effects of anti-platelet drugs and at inhibiting thrombin in the context of ischemia-reperfusion injury of the coronary microcirculation of wildtype and diabetic mice.
It comes some 25 years after the identification of the P2Y12 receptor for ADP by Hollopeter et al.
The authors provide insights into both the inhibition of platelet-rich thrombi formation and the potentially negative effects of increased neutrophil infiltration on cardiac damage and/or repair.
For me, this study emphasizes the need to look again at the molecular interactions that occur during thrombus formation under flow using the modern advances in immunofluorescent imaging as illustrated in the recent manuscript by Kusch et al.
I am thinking not only of primary platelet interactions with themselves and fibrin but also their role in thrombo-inflammation and neutrophil migration across the endothelial barrier into inflamed tissue.
We still have much to learn as this manuscript illustrates before truly optimal personalized combined therapies involving different drugs enter clinical practice. This article by Joanne Mitchell and her coworkers is highly recommended.”
