Shabbir Ansari: Rare F3 Missense Variants Markedly Impair Coagulation Initiation

Shabbir Ansari, Assistant Professor at University of Texas at Tyler School of Medicine, shared on LinkedIn about a recent article he and his colleagues co-authored, published in Blood:

“Excited to share that our new paper in Blood is online!

We demonstrate that selected rare human F3 (tissue factor) missense variants markedly impair coagulation initiation via two distinct mechanisms: disruption of TF–FVIIa binding, or perturbation of a TF exosite required for efficient macromolecular substrate activation (Factors IX and X). Although these variants do not alter standard clinical coagulation assays, carriers exhibit reduced basal coagulation activation in vivo, supporting a previously unappreciated role for TF in supporting basal FVII activation in humans.

As an added bonus, one of the images from this study was selected for the journal’s cover image, recognizing the conceptual and mechanistic insights from this work. Many thanks to my postdoc mentor, Sol Schulman, co-authors, and collaborators for a fantastic team effort.”
