Stefano Navarro: Partial GPVI Downregulation Shows Promise for Safer Antithrombotic Therapy
Stefano Navarro, Postdoctoral Researcher at Rudolf Virchow Center of University of Würzburg, shared a post on LinkedIn about a recent article he and his colleagues co-authored, published in Signal Transduction and Targeted Therapy, adding:
“What if antiplatelet therapy does not always need to completely block a platelet receptor?
This is the idea we explored in our new article in Signal Transduction and Targeted Therapy.
Instead of fully blocking or downregulating the collagen/fibrin (fibrinogen) receptor Glycoprotein (GP) VI, we asked whether its surface expression could be ‘tuned down’ to an intermediate level.
By partially downregulating GPVI, we generated a low-density GPVI phenotype (GPVILO) in which the adhesive function of the receptor was largely preserved, while its intracellular signaling was selectively dampened.
This uncoupling of adhesion from signaling was sufficient to protect from thrombosis and thrombo-inflammation, while preserving hemostasis in our experimental models.
