Muhammad Bilal: Principles and Clinical Uses of the Coombs Test

“Coombs Test (Antiglobulin Test)
1. Objective
The objective of this test was to detect antibodies directed against red blood cells.
2. Principle
The test was based on the reaction between anti-human globulin (Coombs reagent) and antibodies attached to red blood cells. Agglutination indicated a positive result.
3. Materials
- Blood sample
- Coombs reagent (Anti-human globulin)
- Test tubes
- Centrifuge
- Normal saline
- Microscope
4. Procedure
- Blood was collected under aseptic conditions.
- Red blood cells were washed with normal saline.
- Coombs reagent was added to the cell suspension.
- The mixture was centrifuged.
- The tube was examined for agglutination.
5. Result
- A positive result indicates that agglutination was observed.
- A negative result indicates that no agglutination was observed.
6. Uses
- It was used to diagnose hemolytic anemia.
- It helped detect autoimmune reactions.
- It assisted in compatibility testing.
- It was useful in hemolytic disease of the newborn.
7. Consultation
The result was interpreted by a clinician along with clinical findings and other hematological tests.”
