Evangelia Petsalaki: New Systems Biology Approach Identifies Biomarkers in Thrombotic APS
Evangelia Petsalaki, Director of Oncology Data Science Innovation at GSK and Visiting Group Leader at EMBL-EBI, shared on LinkedIn about a recent article she and her colleagues co-authored, published in medRxiv, adding:
”We’re pleased to share our latest preprint on thrombotic primary antiphospholipid syndrome (APS).
In this study, we integrated plasma proteomics, whole-blood transcriptomics, machine learning and network-based analysis to investigate the molecular programmes associated with high-risk disease.
Across multiple complementary analyses, we found evidence for a complement- and interferon-associated thromboinflammatory programme linked to increasing antiphospholipid antibody burden and recurrent thrombosis. Integrating the data using multi-omics factor analysis and network propagation also connected circulating protein signatures to intracellular signalling pathways, providing a systems-level view of disease biology.
Among the findings, Complement Factor D (CFD) emerged as a candidate biomarker associated with recurrent thrombosis, although this will require validation in independent cohorts.
This work was a fantastic collaboration between colleagues at EMBL-EBI, the National and Kapodistrian University of Athens, and the Biomedical Sciences Research Center ‘Alexander Fleming’.
