James Posada: Demonstrating the Benefits of Nuclease Therapy in Ischemic Stroke

James Posada, CEO and Founder of Resolve Therapeutics, LLC, shared a post on LinkedIn:

“Thrombolysis is the standard of care for acute ischemic stroke, unfortunately revascularization is unsuccessful almost 50% of the time.

Two reasons: neutrophil extracellular traps (NETs) make clots resistant to tPA and tenecteplase, and extracellular RNA fuels the inflammation that damages brain tissue once blood flow returns.

This week at the World Stroke Congress in Seoul, we’re presenting in vitro and in vivo preclinical data demonstrating the benefit of nuclease therapy in ischemic stroke.

In a mouse model of ischemic stroke, a single IV dose of our nuclease-Fc fusion proteins (RSLV-132 or RSLV-145) given with tenecteplase:

  • Reduced Day 3 infarct volume by 35% vs. vehicle (p=0.014). TNK alone: 21%, not significant.
  • Preserved motor coordination on the rotarod through Day 3.
  • Restored cerebral perfusion within 2 hours of treatment, which TNK alone did not.

A companion poster shows RSLV-145 dissolving pre-formed NETs within 5 minutes, and both molecules preventing NET formation triggered by RNA-containing immune complexes.

RSLV-132 already has a clean safety record across five clinical trials. We think that makes this a credible path to the clinic for stroke.

Grateful to Gabor Illei, and to Huaxin Sheng and Donovan Edwards at Duke University, for the work behind these posters.
