Alicja Urbaniak: Targeting CD148 to Develop New Antithrombotic Therapies

Alicja Urbaniak, Instructor at UAMS – University of Arkansas for Medical Sciences, Full Member Cancer Therapeutics Program at The Winthrop P. Rockefeller Cancer Institute at UAMS, Chair of the Partnerships Committee at American Society for Pharmacology and Experimental Therapeutics – ASPET, shared a post on LinkedIn about a recent article by Lina El Badaoui et al., published in Pharmacology Research and Perspectives, adding:

“The article ‘Targeting CD148 for Antithrombotic Therapy: Functional and Molecular Evaluation of AKB-9778,’ recently published in Pharmacology Research and Perspectives, explores AKB-9778 (razuprotafib) and its potential to inhibit CD148, a protein tyrosine phosphatase involved in platelet activation.

AKB-9778 partially inhibited thrombus formation in an in vitro whole-blood model. Molecular docking and mutagenesis helped identify residues involved in its interaction with CD148.

However, its potency against CD148 was substantially lower than previously reported, highlighting the need for further characterization and optimization.

These findings suggest that AKB-9778 could serve as a starting point for developing more selective and effective CD148 inhibitors.

While this early-stage research offers a promising direction for antithrombotic drug discovery, clinical efficacy and the potential for a lower bleeding risk remain to be established.⁠”
