A New Approach to Understanding Plasmin Generation and Fibrinolysis – RPTH Journal

”Measuring plasmin generation: a smarter tool for studying fibrinolysis
Fibrinolysis is tightly regulated by plasmin.
Yet reliably measuring plasmin generation in preclinical and clinical settings has remained challenging.
A new study in RPTH Journal presents a novel fluorescence-based plasmin sensor (FPS) designed to address this gap.
Key findings:
- FPS showed higher efficiency for plasmin than existing peptide-based sensors
- Unlike peptide sensors, FPS is not cleaved by plasmin inhibited by α2-macroglobulin giving a more accurate readout
- FPS interacts with kringle domain 5 of plasmin, revealing novel molecular interactions
- When combined with an anti-α2-antiplasmin blocking antibody, FPS can detect nanomolar levels of plasmin
- FPS does not inhibit clot lysis, a key advantage over peptide sensors
- Plasmin generation measured with FPS showed only 2-fold variation across healthy volunteers — suggesting good reproducibility
- The sensor can also detect the influence of PAI-1 when antiplasmin is inhibited
Better tools to measure plasmin generation could significantly advance our understanding of fibrinolytic disorders and ultimately improve how we diagnose and manage conditions like hyperfibrinolysis or thrombosis.”
