Jagadeesh Bayry: Molecular and Genetic Mechanisms of VITT
Jagadeesh Bayry, Professor and Head of Department at the Indian Institute of Technology Palakkad, shared a post on LinkedIn and about a recent article he and his colleagues co-authored, adding:
“An editorial commentary in Nature Reviews Cardiology, Nature Portfolio, co-authored with Julie Rayes, discusses the mechanisms underlying Vaccine-induced immune thrombotic thrombocytopenia, a rare complication of adenoviral vector-based COVID-19 vaccines.
An elegant study by the groups of J. J. Wang and Andreas Greinacher demonstrates that VITT arises from molecular mimicry between platelet factor 4 PF4 and adenoviral core protein VII.
In genetically predisposed individuals carrying IGLV3-21 star 02 or star 03 alleles, a key somatic mutation K31E in the IgG light chain redirects antiviral immunity toward pathogenic anti PF4 responses, triggering platelet activation and thrombosis.
This work provides a compelling example of how genetic background and somatic hypermutation converge to drive autoimmunity, and opens new avenues for designing safer adenoviral vector platforms.”
