Gurpreet Kaur Sagoo: HPLC vs HbCZE in Comparison of HbA2
Gurpreet Kaur Sagoo, Professor at Armed Forces Medical College, shared a post on LinkedIn about a recent article she and her colleagues co-authored, published in Indian Journal of Hematology and Blood Transfusion, adding:
“High Performance Liquid Chromatography (HPLC) and Haemoglobin Capillary Zone Electrophoresis (HbCZE) are primary automated tools for hemoglobinopathy diagnosis, identifying variants and quantifying HbA2/HbF for thalassemia screening.
Method Principles
HPLC uses cation-exchange chromatography to separate hemoglobins by retention time, excelling in routine quantification of HbA2 and HbF. HbCZE separates by electrophoretic mobility in alkaline buffer across 15 zones, aiding variant migration identification.
Diagnostic Performance
They reliably detect common variants (HbD-Punjab, E, C, S/A, S/S) but differ in HbA2 accuracy:
- HPLC underestimates in HbD/E traits due to co-elution
- Overestimates in HbS
- HbCZE provides better resolution but overlaps in HbC/S/D cases.”
