Alan Nurden: A New Mechanism Behind Immune Thrombocytopenia

Alan Nurden, Emeritus Research Director at CNRS, Co-Founder of the French National Reference Centre for Inherited Platelet Disorders (CRPP), shared a post on LinkedIn about a recent article by Jiaqian Qi et al., published in Nature Communications, adding:

“For over 50 years, Changgeng Ruan established Suzhou in China as a worldwide center for studies not only on platelets but also many other aspects of hematology.

Now it is clear that the future in Suzhou is in good hands. In this paper published in Nature Communications, Yue Han and her colleagues present a new hypothesis for the pathogenesis of immune thrombocytopenia (ITP) in which neutrophil-derived alarmins, and in particular S100A8/9, interfere with megakaryocyte maturation and migration in the bone marrow; in so doing they have a negative influence on thrombopoiesis and platelet production.

The authors provide several lines of evidence to support this, including a restoration of platelet numbers by pharmacological blockade.

This is potentially a major paper and is highly recommended.

The relationship with the antiplatelet antibodies supposed to be at the origin of ITP remains to be elucidated, do they remain as the cause or are they formed secondarily to the action of the alarmins.”
