Jennifer Trowbridge: How Mutant Blood Stem Cells Use Inflammation to Drive Their Own Expansion

Jennifer Trowbridge, Professor, Dattels Family Endowed Chair and Chair of Stem Cells and Development Program at The Jackson Laboratory, shared a post on LinkedIn about a recent article she and her colleagues co-authored, published in Haematologica, adding:

“Excited to share that new work from our lab, led by Jayna Mistry, is now published in Haematologica Journal!

How do mutant blood stem cells create an inflammatory environment that favors their own expansion?

We found that DNMT3A-mutant neutrophils produce elevated S100A8/A9, promoting inflammatory signals including TNFα and IL-6.

Targeting S100A8/A9 with either tasquinimod or a neutralizing antibody reduced mutant hematopoietic stem cell expansion in mouse models. These findings dissect a feed-forward loop linking inflammation and clonal hematopoiesis, and suggest an intervention strategy to target it.

Grateful to our funding sources National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK), National Institute on Aging (NIA), The Edward P. Evans Foundation and Blood Cancer United and collaborators Rongbao Zhao and Amit Verma!”
