Ron DePinho: A Negative Trial Does Not Repudiate Inflammation as a Cardiovascular Target
Ron DePinho, Professor at Department of Cancer Biology, Harry Graves Burkhart III Distinguished University Chair Cancer Biology at MD Anderson Cancer Center, shared a post on LinkedIn:
“Disappointing for this program, but definitely not a repudiation of inflammation as a cardiovascular target.
The key question will be whether the trial tested the right inflammatory biology, in the right patients, at the right stage of disease.
Cardiovascular inflammation is not a single pathway or biomarker; it is a dynamic network shaped by residual lipid risk, obesity and metabolic dysfunction, clonal hematopoiesis, immune aging, and tissue injury.
The field should learn as much from a negative trial as from a positive one. We need better mechanistic patient stratification, pharmacodynamic biomarkers that demonstrate pathway engagement in the relevant tissue, and combination strategies matched to the biology of residual risk.
The inflammatory hypothesis remains compelling in my view … it is likely that its clinical translation clearly requires greater precision.”
