”One sample… two methods… completely different reliability levels

Slide method

Simple and rapid – results in just 5–10 minutes
Great for emergency field blood typing
Uses small amounts of reagents
Low sensitivity – easily affected by multiple variables
Cannot reliably detect weakly expressed or rare antigens
False negatives can occur with low-titer anti-A or anti-B antibodies – a serious transfusion risk!
Drying of the slide can mimic agglutination (false positive!)

Tube Method

Considered the gold standart in blood bank serology
Uses centrifugation to enhance antigen–antibody reactions – even weak reactions are detected
Supports both forward (cell) and reverse (serum) grouping
Longer incubation is possible without sample drying
More reliable for crossmatching before transfusion
Takes more time and requires more equipment
Needs a centrifuge – not always available in resource-limited settings
