Bilal Salman։ NETosis as a Therapeutic Target in Inflammation and Thrombosis
Bilal Salman, Student at University of Balamand, shared a post on LinkedIn about a recent article he and his colleagues co-authored, published in European Journal of Inflammation, adding:
”Neutrophil extracellular traps (NETs) are increasingly recognized as important mediators linking innate immunity, inflammation, thrombosis, and cardiovascular remodeling.
These figures from our most recent publication highlight potential therapeutic targets involved in ROS generation (Apocynin, DPI, Colchicine, Thiocyanate, Selenocyanate), ROS clearance (GSH + NAC), NET formation (GSK), NET clearance (DNase I), as well as immunomodulatory therapies (Anakinra, IL-1 monoclonal antibody, and anti-IL6R therapy).
Targeting ROS may reduce the signals that initiate NETosis. Targeting PAD4 may prevent NET formation directly, while DNase I can help remove NETs after they have formed. Immunomodulators such as anakinra, IL-1β blockade, and anti-IL-6R therapy instead target the inflammatory environment surrounding NETosis.
The major challenge is finding the right balance between suppressing pathological NETosis and preserving the essential antimicrobial functions of neutrophils.
NETosis therefore represents a fascinating therapeutic intersection between oxidative stress, innate immunity, inflammation, thrombosis, and cardiovascular disease.”
