James P. Crowley: A New Adult Progenitor with Erythro-Myeloid Potential
James P. Crowley, Professor of Medicine emeritus at Brown University, shared a post on LinkedIn about a recent article by Shweta Tikoo et al., published in Nature Immunology, adding:
“One of the central tenets of macrophage development in the adult is their monocytic origin in inflammation and cancer.
Whether additional progenitor lineages contribute toward macrophage production in adulthood is unclear.
Here they identify and characterize a previously unrecognized adult BM progenitor, EMPROR, with robust erythro–myeloid (EMP) potential.
The discovery of EMPROR (CD4-lineage Erythro-Myeloid Progenitor Residing in the Organism’s Reservoirs) cells fundamentally challenges this paradigm. EMPROR cells are distinct from cMoPs and monocytes by phenotypic marker expression, transcriptional state and lineage potential in vitro and in vivo.
Unlike cMoPs and monocytes, EMPROR cells lack DC potential but generate erythroid, neutrophil, megakaryocytic and macrophage lineages.
These findings raise the possibility that macrophage replacement or augmentation in injured or inflamed tissues may, under some circumstances, be driven by precursors with an EMP-like program rather than solely by monocyte recruitment.
A central conceptual implication of these findings is that an embryonic erythro–myeloid program, long thought to be developmentally restricted, persists into adulthood.”
