Pablo Corral: Is Inflammation Really a Therapeutic Target in Atherosclerosis
Pablo Corral, Pharmacology Professor at FASTA University, Past President of Argentine Lipid Society, shared a post on LinkedIn about a recent article by Kevin Jon Williams, published in JIM, adding:
“Anti-inflammatory agents in atherosclerosis – and a need for reform: Extraordinary claims require extraordinary evidence
A provocative and timely Perspective by Kevin Jon Williams challenging the inflammatory hypothesis of atherosclerosis.
- ZEUS adds another null result: targeting inflammation did not translate into fewer cardiovascular events (HR approximately 0.99).
- Williams highlights a much broader issue: more than 50 clinical trials of anti-inflammatory strategies in atherosclerosis over decades have produced a remarkably disappointing clinical track record.
- His challenge to the field is uncomfortable but important: should inflammation continue to be considered a definitive therapeutic target in atherosclerosis when repeated clinical testing has failed to deliver consistent benefit?
- In sharp contrast, lowering cholesterol-rich ApoB-containing lipoproteins rests on extensive and reproducible evidence targeting the proven causal agents of atherosclerosis.
As evidence accumulates, are we witnessing regression to the mean – or regression to the truth?
Inflammation may be part of atherosclerosis – but being part of the disease does not make it a proven therapeutic target.”
