Shehab Mohamed: Exploring the Emerging Link Between JAK2 Mutations and Thoracic Aortic Aneurysms

We are pleased to share our newly published review exploring the emerging association between JAK2 mutations and thoracic aortic aneurysms in patients with myeloproliferative neoplasms and clonal hematopoiesis.

Title: Association between JAK2 Mutations and Thoracic Aortic Aneurysms in Myeloproliferative Neoplasms

This review summarizes current clinical and mechanistic evidence linking JAK2 V617F mutations with thoracic aortic aneurysm formation, highlighting the evolving role of clonal hematopoiesis beyond hematologic malignancy and thrombosis.

Across multiple cohorts involving more than 446,000 participants, JAK2 positivity was consistently associated with increased thoracic – but not abdominal – aortic aneurysm risk.

We also discuss the potential biologic mechanisms underlying this association, including inflammatory cytokine signaling, macrophage activation, endothelial dysfunction, extracellular matrix degradation, and vascular remodeling driven by JAK-STAT pathway activation.

Importantly, this work emphasizes the growing intersection between hematology, cardiovascular disease, inflammation, and precision medicine, while highlighting the need for prospective studies to determine the clinical implications of vascular surveillance in JAK2-mutated patients.

This publication reflects our continued interest in myeloid neoplasms, clonal hematopoiesis, thrombosis, and translational hematology research.
