Mohammad Reza Moghaddasnejad: Platelet Activation as a Key Driver of Breast Cancer Progression

“Breast cancer cells alter platelet biology.
Breast cancer cells influence platelet biology through multiple mechanisms.
These cells secrete interleukin-6 (IL-6), which stimulates the liver to increase the production of thrombopoietin (TPO).
Elevated levels of TPO promote megakaryocyte maturation in the bone marrow, leading to an increased number of circulating platelets in patients.
In addition, tumor cells release various soluble factors and exosomes that act on peripheral blood platelets, altering their transcriptome and proteome.
These alterations generate a platelet phenotype that actively contributes to the progression of breast cancer.
Moreover, breast cancer cells participate in platelet activation by directly interacting with platelets and secreting tissue factor (TF), which induces thrombin generation and facilitates the platelet activation process.
In their direct interaction with platelets, breast cancer cells express surface ligands such as podoplanin, mucin, CD44, and CD97, which bind to their respective receptors on platelets—including CLEC-2, CD62P, and integrin αIIbβ3—and play key roles in triggering platelet activation.”
