Jim Vadolas: New Evidence Linking Iron Dysregulation to Inflammation in Myelodysplastic Syndromes
Jim Vadolas, Group Leader at Hudson Institute of Medical Research, shared a post on LinkedIn about a recent article by Shahla Vilcassim et al. published in American Journal of Hematology, adding:
“Proud to share our latest publication in the American Journal of Hematology:
‘Therapeutic Silencing of Tmprss6 Reduces Iron-Induced Inflammation and Prolongs Survival in MDS Mice’
In this study, we investigated how dysregulated iron homeostasis contributes to inflammation in myelodysplastic syndromes (MDS) using the NHD13 murine model.
We demonstrate that hepatocyte-targeted silencing of TMPRSS6 increases hepcidin expression, redistributes iron, suppresses inflammasome activation, delays haematological decline and prolongs survival.
These findings provide mechanistic evidence linking iron metabolism to innate immune activation in MDS and establish proof-of-concept that therapeutic modulation of iron homeostasis may represent a disease-modifying strategy for bone marrow failure disorders.
This work represents a truly collaborative effort.
I would like to acknowledge the outstanding contributions of first author Shahla Vilcassim and George Grigoriadis and our research team, including Nuttanan Pholngam, Rattanawan Thubthed, Tiwaporn Nualkaew, Saovaros Svasti, Pornthip Chaichompoo (Mahidol University), Kai Kysenius, Peter J. Crouch (The University ofMelbourne), Sibylle Dames, Mona Eisermann, Alberto Martinez, and Ute Schaeper (Silence Therapeutics).
We are also grateful for the support of Hudson Institute of Medical Research, mRNA Victoria Research Acceleration Fund (mVRAF), and Silence Therapeutics, whose contributions were instrumental to the completion of this work.”
