Alan Nurden: How Platelets Construct a Thrombus Under Flow
Alan Nurden, Emeritus Research Director at CNRS, Co-Founder of the French National Reference Centre for Inherited Platelet Disorders (CRPP), shared a post on LinkedIn about a recent article by Robert H. Lee et al., published in Blood, adding:
”This short communication in Blood from Robert Lee and his colleagues contains a wealth of information on how platelets construct a thrombus under flow.
Combining studies on human hypofibrinogenemia with up-to-date mouse models that reproduce the disease state and/or an altered Fg αC region function, they concluded that the latter regulated arterial thrombosis under hypofibrinogenemic conditions.
More generally, they showed that fibrin-dependent thrombus construction involved both GPVI dependent and independent pathways.
Given that the ISTH Subcommittee on Health and Disease is currently surveying the very rare occurrence of arterial thrombosis in patients with Glanzmann thrombasthenia, themselves already protected by the absence of alphaIIbbeta3 (or with nonfunctional alphaIIbbeta3), looking at the fibrinogen involvement in their rare thrombotic events (both arterial and venous) may well offer some clues.
It is amazing how such natural pathophysiological variants selectively affecting one or more aspects of thrombus formation, either platelet borne or affecting coagulation pathways, are providing essential information towards the construction of therapies better able to combat cardiovascular disease in the general population.”
