Swarnava Roy: Linking Angiotensin Signaling to Oxidative Stress and Organ Damage in Sickle Cell Anemia

“My Recent Publication in JCI Insight!
I am thrilled to share my latest article, ‘Angiotensin signaling is essential for stress erythropoiesis but causes retention of dysfunctional mitochondria in RBCs,’ recently published in JCI Insight!
This work was a wonderful collaborative effort at Cincinnati Children’s Hospital with my brilliant co-authors: Parul Rai, Paritha Arumugam, Diamantis G. Konstantinidis, Sithara Raju Ponny, Marthe-Sandrine Eiymo Mwa Mpollo, Archana Shrestha, Theodosia A. Kalfa, and Punam Malik.
Key Findings:
- The Vicious Cycle: We discovered that in sickle cell anemia (SCA), erythroid-generated reactive oxygen species (ROS) create a dangerous positive feedback loop with Angiotensin II ({AT}—{ATR1}) signaling.
- Mitochondrial Retention: While this signaling is essential for stress erythropoiesis, it unfortunately causes mature red blood cells to retain dysfunctional mitochondria, fueling further ROS production.
- End-Organ Damage: This retention perpetuates hyperangiotensinemia, leading to severe complications like SCA-associated nephropathy. Notably, blocking this pathway successfully broke the cycle and reduced tissue damage.
These insights pave the way for exciting new therapeutic strategies to protect patients from devastating organ damage.
