Joshua Muia: The Evolution of ADAMTS13 Research

Joshua Muia, Associate Investigator at Versiti Blood Research Institute, shared a post on LinkedIn about a recent article he and his colleagues co-authored, published in Thrombosis Update, adding:

“I am pleased to share our invited review, ‘ADAMTS13 at 25: From the Mystery of Thrombotic Thrombocytopenic Purpura to the Molecular Paradigm of Mechanoregulated Proteolysis.’

This article is part of a special issue celebrating the 5th anniversary of Thrombosis Update and commemorating 25 years since the discovery of ADAMTS13, a breakthrough that transformed our understanding of thrombotic thrombocytopenic purpura (TTP).

In this review, we examine:

  • The historical discoveries that led to the identification of ADAMTS13.
  • How mechanical forces regulate von Willebrand factor (VWF) cleavage by ADAMTS13.
  • Emerging concepts of allosteric regulation, autoinhibition, and global latency.
  • Advances in diagnostics, biomarkers, and targeted therapies for TTP.
  • Future opportunities in structural biology, precision medicine, and ADAMTS13 engineering.

Many thanks to my co-authors and mentees Olamilekan George, Thomas Momanyi, Frida Miranda, and everyone who has advanced ADAMTS13 research for their contributions that continue to make an impact on the field.”
