Dhiren Patel: PLD1-Derived Phospholipids Regulate NAD Kinase and ROS Production in Neutrophils
Dhiren Patel, Postdoctoral Researcher at Max Planck Institute for Infection Biology, shared a post on LinkedIn about a recent article he and his colleagues co-authored, published in Nature, adding:
“Delighted to share my first independent research project with Arturo Zychlinsky at Max Planck Institute for Infection Biology now available in Nature Communications.
In Rahaf Albarghash’s Master’s project we demonstrate that NAD kinase is regulated by Phospholipase D1-derived phospholipids in neutrophils.
Shotgun lipidomics and curated inhibitor screens revealed that the enzyme phospholipase D1 (PLD1) triggers the production of the phospholipids phosphatidic acid and PIP₂.
PLD1 also promotes reactive oxygen species production in response to numerous neutrophil extracellular trap and phagocytic stimuli.
These results were confirmed genetically using PLD1 knockouts in CD34⁺-derived neutrophils generated by Sergio G. San Miguel. More to come from him soon.
NAD kinase activity was thought to be regulated by phosphorylation.
We demonstrate that PLD1-derived phospholipids can bind and boost the activity of NAD kinase, promoting the NADPH supply needed for reactive oxygen species generation.
NAD kinase is the only mammalian enzyme that directly synthesises NADP⁺, so these findings may extend beyond neutrophil function and into other immune cells, and particularly tumour metabolism.”
