Monica Schaller: How Do Autophagy, Inflammation, and NETs Interact to Drive Autoimmune Thrombosis?

Monica Schaller, Assistant Professor, Coordinator of Graduate School for Cellular and Biomedical Sciences (GCB), at University of Bern, shared on LinkedIn about a recent article she and her colleagues co-authored, published in Frontiers, adding:

”How do autophagy, inflammation, and neutrophil extracellular traps (NETs) interact to drive autoimmune thrombosis?

Our newly published review explores this fascinating and complex crosstalk and its implications for diseases for three important thrombotic autoimmune disorders:

  • Immune Thrombocytopenia (ITP)
  • Heparin-Induced Thrombocytopenia (HIT)
  • Immune-mediated Thrombotic Thrombocytopenic Purpura (iTTP)

Over the past years, growing evidence has highlighted the central role of NETs and inflammatory pathways in autoimmune diseases.

In this review, we bring together current knowledge and propose a conceptual framework in which autophagy, NET formation, and inflammation form a self-reinforcing network that contributes to immunothrombosis and disease progression (Figure 1, below).

Our review discusses how dysregulated NET formation may promote thrombosis, autoimmunity, and endothelial injury, and explores whether targeting autophagy and NET-associated pathways could offer novel therapeutic opportunities in these challenging diseases.
