Jae Hyun Byun: Beyond Stenosis Understanding Carotid Plaque Instability

Jae Hyun Byun, PhD Doctoral Research Trainee at Vascular Health Unit, shared a post on LinkedIn about a recent article he and his colleagues co authored, published in ESC, adding;

“Carotid plaque instability is a major cause of ischemic stroke, yet current clinical guidelines rely heavily on stenosis severity, which may not fully capture the underlying disease biology.

I’m excited to share our latest research published in Cardiovascular Research!

In this study, we applied CosMx whole transcriptome single cell spatial transcriptomics on human carotid atherosclerotic plaques, profiling 18,934 RNA targets at single cell and subcellular resolution.

By integrating single cell RNA sequencing of carotid plaques and paired peripheral blood mononuclear cells from the same patients with spatial transcriptomics, we reveal distinct vascular and immune microenvironments associated with plaque instability, highlighting the importance of assessing beyond stenosis severity.

A huge thank you to the Vascular Health Unit team and my supervisor Stella S. Daskalopoulou (She Her), our collaborators at the McGill Genome Centre (Ioannis (Jiannis) Ragoussis, Ashot Harutyunyan, Spyros Oikonomopoulos), the RI MUHC histology core (Fazila C.) and Bruker Spatial Biology (Joseph Beechem,Joseph Beechem, Kate E., Ben Landas) for their incredible support and contributions!”
