Eugene Bolotin: Thrombosis and Survival in Pancreatic Cancer

Eugene Bolotin, Chief Executive Officer at MiraOmics, shared a post on LinkedIn:

“Back to the horrors of pancreatic cancer this time, thrombosis.

Pancreatic cancer creates a dangerous tendency toward abnormal blood clotting.

Thrombosis means a blood clot forms inside a blood vessel. Complications include clots in the legs, lungs and abdominal vessels, as well as heart attacks and strokes.

One prospective study found venous clotting in about one in five patients; another found arterial events in nearly 6% of older patients within six months.

The cancer specific clotting biology remains incompletely understood.

We screened 152 TCGA pancreatic cancer slides from 130 patients for thrombus like candidate regions in routine H&E.

Overall survival data were available for 128 patients.

The figure shows model flagged examples, hotspot count maps and overall survival.

Higher candidate counts tracked shorter survival in an exploratory comparison (unadjusted p  equal 0.012).

The cutpoints were chosen in this cohort.

NCI recognized this as a high priority and is already funding a 3.1 million dollar project on pancreatic cancer’s coagulation biology.

Hopefully, this effort will shed some light on the mechanism of this complications.

I want to thank and credit Samarpan Majumder one of the recipients of the NCI grant for discussion that inspired this analysis.
