Swarnava Roy: Key Findings on Stress Erythropoiesis and Mitochondrial Retention in RBCs

“My Recent Publication in JCI Insight!
I am thrilled to share my latest article, ‘Angiotensin signaling is essential for stress erythropoiesis but causes retention of dysfunctional mitochondria in RBCs,’ recently published in JCI Insight!
This work was a wonderful collaborative effort at Cincinnati Children’s Hospital with my brilliant co-authors: Parul Rai, Paritha Arumugam, Diamantis G. Konstantinidis, Sithara Raju Ponny, Marthe-Sandrine Eiymo Mwa Mpollo, Archana Shrestha, Theodosia A. Kalfa, and Punam Malik.
Key Findings:
- The Vicious Cycle: We discovered that in sickle cell anemia (SCA), erythroid-generated reactive oxygen species (ROS) create a dangerous positive feedback loop with Angiotensin II-mediated AT1 receptor (AT1R) signaling.
- Mitochondrial Retention: While this signaling is essential for stress erythropoiesis, it unfortunately causes mature red blood cells to retain dysfunctional mitochondria, fueling further ROS production.
- End-Organ Damage: This retention perpetuates hyperangiotensinemia, leading to severe complications like SCA-associated nephropathy.
Notably, blocking this pathway successfully broke the cycle and reduced tissue damage.
