Rupam Manna: CELMoDs – Next-Generation Molecular-Glue Protein Degraders
Rupam Manna, Postdoctoral Student – DrNB Medical Oncology at National Board of Examinations, shared a post on X:
“CELMoDs: next-generation molecular-glue protein degraders.
By binding cereblon, CELMoDs redirect the CRL4–CRBN E3 ubiquitin ligase toward IKZF1 and IKZF3.
Their proteasomal degradation suppresses IRF4–MYC signalling, promotes myeloma-cell apoptosis and activates T and NK cells.
Key agents:
- Iberdomide – first FDA-approved CELMoD
- Mezigdomide – investigational in multiple myeloma
- Golcadomide – investigational in lymphoma
Remember the major class toxicities: neutropenia, infections, anaemia and thrombocytopenia.”
