Abdurrahman Tufan: Combination Targeted Therapy in Refractory Systemic Autoinflammatory Diseases

“I’m very pleased to share our new publication in Seminars in Arthritis and Rheumatism:
‘Biologic biologic and biologic JAK inhibitor combination therapy in refractory systemic autoinflammatory diseases.’
Our findings demonstrate the potential of combination targeted therapy to achieve sustained disease control in selected patients with severe and refractory systemic autoinflammatory diseases (SAIDs), including those with persistent inflammation and limited therapeutic options.
In this retrospective longitudinal cohort from the NIH Inflammatory Disease Section, we evaluated advanced combination therapy (ACT) using biologics and or JAK inhibitors in patients with difficult to treat SAIDs.
Among 38 patients, 65 ACT regimens were administered. Following multiple treatment trials:
- 55.3 percent achieved a complete response
- 31.6 percent achieved a partial response
- 68 percent of patients remained on combination therapy at final follow up
- Median duration of ACT was 5 years (range 11 to 186 months)
